How to use biological safety cabinet

1. Installation of the instrument: The installation requirements of different types of biological safety cabinets are different and should be strictly implemented in accordance with the installation requirements of the safety cabinets. (1) During the handling of the biological safety cabinet, it is strictly prohibited to lay it down and disassemble it. It is advisable to unpack it after moving into the installation site. (2) The biological safety cabinet should be installed near the exhaust vent, should not be installed in the place where the airflow changes drastically and people move a lot, and should not be installed at the door. The biological safety cabinet should be downstream of the airflow direction. (3) The distance between the back and sides of the biological safety cabinet and the wall should be maintained at an inspection distance of not less than 150㎜ ~ 300㎜, and there should be at least 300㎜ space on the top. (4) The exhaust system should be able to ensure that the biological safety cabinet is under negative pressure relative to the room where it is located, and it should be noted that the installation of the exhaust air HEPA filter must be replaced when installing. (5) If other gases are needed in the safety cabinet, gas pipes should be installed at the same time. 2. Precautions for experiment operation in the biological safety cabinet: (1) Place the items in the cabinet in parallel; (2) The operation should be slow; (3) Avoid cross-contamination when moving the items in the cabinet; (4) Avoid vibration; (5) ) Try not to use an open flame in the cabinet. 3. Maintenance and care: (1) The studio should be cleaned and disinfected after each experiment. (2) Once the pre-filter is damaged, it should be replaced in time. (3) After the service life of the HEPA filter expires, it should be replaced immediately. Since the filter may contain contaminants, attention should be paid to safety protection during operation, and it is best to ask a professionally trained professional to replace it. (4) The biosafety cabinet should be tested on-site when one of the following situations occurs: ①After the completion of the biosafety laboratory and before it is put into use, the biosafety cabinet has been installed; ②The location of the biosafety cabinet has been moved; Overhaul; ④ After replacing the HEPA filter in the biological safety cabinet; ⑤ Annual routine inspection of the biological safety cabinet. Test items include vertical airflow speed, working window airflow direction and flow rate, cleanliness of work area, noise, light intensity, exhaust HEPA filter leak detection, etc. The detection should meet certain requirements: ①Vertical airflow average wind speed detection detection method: filtering in HEPA On the section at 0.15m below the device, the wind speed is measured uniformly by using an anemometer. The distance between measuring points is not more than 0.15m, at least 3 points are measured in each column, and at least 5 points are measured in each row. Evaluation standard: The average wind speed is not less than 0.25m / s, the difference from the manufacturer's given value is not more than ± 0.025m / s, and the difference between the single-point wind speed and the average wind speed is not more than ± 20%. ②The airflow direction of the working window can be detected on the cross section of the working window using the smoke method or the silk method. The detection position includes the surrounding edges and the middle area of the working window. Evaluation standard: the airflow at all positions of the working window section is inward. ③The detection method of the airflow velocity of the working window: use an anemometer to measure the wind speed of the working window section. The distance between measuring points is not more than 0.10m, at least 2 points are measured in each column, and at least 5 points are measured in each row. Evaluation standard: The wind speed value on its cross section is not lower than the product standard requirements. ④The cleanliness of the work area is detected by a dust particle counter in the work area. The sampling port of the particle counter is placed at a height of 20 cm above the table, and its measurement points follow a grid distribution of 20 cm in rows and columns. Measure at least three points in each column and at least 5 points in each row. Evaluation standard: The number of dust particles should not exceed the requirements of product standards. ⑤ Noise detection The front center of the front panel of the biological safety cabinet is 300㎜ outwards, and 380㎜ above the work surface. Use a sound level meter to measure noise. Evaluation standard: The noise should not be higher than the product standard requirements. ⑥ Illumination detection Set a measuring point every 30cm along the center line of the length of the worktable. When the distance from the side wall is less than 15 cm, it is no longer set. Evaluation standard: The average illuminance is not lower than the product standard requirements. ⑦ Leakage detection of the cabinet should be carried out when conditions permit. The biosafety cabinet can be sealed and pressurized to a pressure of 500 Pa to detect leaks with soap bubbles.
